Goedkoper én veiliger: De groene revolutie in het Vietnamese chemielokaal

Stel je voor! Een bruisend chemielabo waar studenten de geheimen van moleculen ontrafelen, zonder risico om blootgesteld te worden aan giftige dampen of kankerverwekkende stoffen. Hoewel veel practica nog steunen op verouderde experimenten en gevaarlijke chemicaliën, bewijst onderzoeker Jenno Vanassche (Universiteit Antwerpen) in zijn scriptie dat het anders kan. Hij bedacht een systematische methode om proeven met gevaarlijke chemicaliën te vervangen door veiligere, goedkopere en groenere alternatieven. Het grote pluspunt? De studenten leren er exact evenveel van en lopen geen praktijkervaring mis.


Organische chemie in de praktijk: essentieel maar risicovol

Organische chemie is de wetenschap van koolstofverbindingen, de bouwstenen van vrijwel alles om ons heen. Praktijkproeven zijn voor studenten hier dé manier om chemie in de vingers te krijgen, maar aan de Quy Nhon Universiteit (QNU) in Vietnam zorgt dit soms voor hachelijke situaties. Een aantal experimenten steunen er nog op gevaarlijke chemicaliën die giftig, kankerverwekkend, extreem brandbaar of schadelijk voor het milieu zijn. Waar Westerse laboratoria deze risico's indijken met talrijke zuurkasten en dure afvalverwerking, is de infrastructuur in Quy Nhon vaak een stuk beperkter. 

Hierdoor is de kans op blootstelling voor studenten groter en wordt het lokale milieu zwaarder belast. Toch is er licht aan het einde van de tunnel: het onderzoek toont aan voor een aantal chemieproeven veiligere alternatieven bestaan zonder in te boeten naar het leerdoel. Door slimme, groene alternatieven te omarmen, krijgt de Vietnamese universiteit een tastbare oplossing in handen om haar laboratoria veiliger en toekomstklaar te maken.

Van gevarenlijst naar echte vervanging

De makkelijkste oplossing lijkt simpel: schrap de gevaarlijke proeven gewoon. Maar dat is geen optie, want dan missen de studenten essentiële praktijkervaring. De beste weg vooruit is daarom substitutie: het gericht vervangen van de chemische boosdoeners door veiligere alternatieven. Toch is dat een flinke uitdaging, want een stof zit niet zomaar in een experiment; ze dóét iets. Een oplosmiddel zorgt ervoor dat stoffen bijvoorbeeld mengen of verdund worden en een reagens lokt de eigenlijke reactie uit. Je kan dus niet zomaar een lijstje met "gevaarlijk" en "veilig" afvinken, want de vervangende stof moet de proef nog altijd laten slagen.

"Vergelijk het met het bakken van een taart: als je eieren wilt weglaten omdat iemand allergisch is, moet je een alternatief zoeken dat de taart nog steeds luchtig houdt. Een vervangende stof moet nog tot het gewenste product leiden, anders mislukt de proef en leren de studenten niets."

Hiervoor vertrok Vanassche vanuit een bestaand digitaal EHS-instrument (Environment, Health and Safety). Daarbij kreeg eerst elke afzonderlijke stof in een experiment een eigen veiligheidscore op basis van strikte criteria, zoals brandbaarheid, corrosiviteit, giftigheid, kankerverwekkend en milieuschade. Al deze individuele stofscores werden vervolgens samengevoegd tot één globale 'gevaarscore' voor het volledige experiment. Zo werd meteen duidelijk welke proeven in hun totaliteit het meest problematisch waren. 

Aan deze globale gevarenscan werd een cruciale 'functionele analyse' toegevoegd. Hierbij keek de onderzoeker naar de precieze rol van de stof in de reactie. Voor oplosmiddelen werd bijvoorbeeld de zogenaamde Hansen methode gebruikt om te berekenen of de alternatieve vloeistoffen wel dezelfde oploskracht bezitten. Uiteindelijk werden de kandidaat-vervangers streng met elkaar vergeleken op basis van een slimme balans: het moet de gevaarscore van het experiment stevig naar beneden halen, een haalbare kostprijs hebben, én zorgen voor een hoge reactie-efficiëntie zodat er zo min mogelijk chemisch afval overblijft.


Drie proeven, drie verschillende oplossingen

Uit de analyse filterde Vanassche een top 10 van de meest risicovolle experimenten, geranschikt volgens gevarenscore. Drie daarvan nam hij grondig onder de loep om te bewijzen dat gerichte vervanging werkt (figuur 1).

 

1. Aniline: veiliger én efficiënter

Bij de bromering van aniline scoorde het origineel experiment zeer hoog (9,8), voornamelijk vanwege het gebruik van het toxische aniline en corrosief 𝐵𝑟2. Door aniline te vervangen door het veiligere acetanilide en gebruik te maken van een alternatief bromeringsreagens (KBr, waterstofperoxide en azijnzuur), werd de gevarenscore teruggebracht tot 5,5 en de atoomeconomie verhoogd van 57,6% naar 74,3%. Een andere, nog goedkopere route (met NaBr, NaOCl en HCl) verlaagde de gevarenscore eveneens aanzienlijk ten opzichte van het origineel, wat aantoont dat veiligheid en betaalbaarheid uitstekend hand in hand kunnen gaan.

 

2. Aceton: slimmer omgaan met oplosmiddelen 

De aanmaak van traanverwekkend broomaceton-experiment (gevaarscore 9,4) gebruikte het ozonaantastende oplosmiddel CCl₄ in combinatie met giftig broom. Naast het gebruik van een milieuvriendelijker solvent, (bv. propyleencarbonaat) kan door over te schakelen op HBr en H₂O₂ het actieve broom veilig in de kolf worden aangemaakt. Deze gebalanceerde route heeft een hoge atoomeconomie (79,2%) en kost drie keer minder dan het originele experiment.

 

3. Harsen: van giftige cocktail naar dennenextract 

De productie van fenol-formaldehydehars gebruikt een giftige cocktail van uiterst giftige stoffen (score 9,0) zoals formaldehyde en fenol. Vanassche stelde een biogebaseerd alternatief voor: een mix van 'tannine' (een natuurlijk plantenextract) en glyoxal. De gevaarlijkheid daalt drastisch (score 5,5) en het is tevens goedkoper. Wel moet de vorming van dit complexere hars nog in het labo gevalideerd worden.

Meer dan een Excelbestand: de volgende stap 

De kern van dit onderzoek is niet zomaar het zoeken naar één nieuwe chemische stof, maar een fundamentele manier van denken met groene chemie principes voorop. Eerst worden de gevaarlijkste proeven gevonden, dan wordt naar de echte functie van de gebruikte chemicaliën gezocht, waarna eliminatie, vervanging of herontwerp volgt. Hoewel vervanging vaak mogelijk is, werden sommige proeven ook vervangen door een demonstratievideo als dat het veiligste leerpad bleek. 

De resultaten vormen een krachtig, toekomstgericht vertrekpunt naar groenere chemie onderwijs in QNU. De bevindingen bewijzen dat we chemielessen fundamenteel veiliger en milieuvriendelijker kunnen maken, zelfs met beperkte middelen. Veiliger chemieonderwijs begint dus niet met minder chemie, maar met slimmer kiezen en tegelijk de principes van groene chemie aanleren en dat is, letterlijk en figuurlijk, een verademing.
