Metagenomic study of lettuce potting soil microbiome under chitin treatment and functional genomic analysis of Mortierella hyalina

Een wereld zonder pesticiden?

Voorspellingen van de Verenigde Naties schatten dat er tegen het jaar 2050 bijna 10 miljard mensen op aarde zullen leven, waarvan de meesten in derdewereldlanden zullen worden geboren. Het is geen simpele opdracht om al deze mensen op een duurzame manier van voeding te voorzien. Om een zekere kwaliteitsstandaard voor hun gewassen te garanderen maken landbouwers gebruik van chemische additieven, namelijk kunstmest en pesticiden, wat een negatieve impact kan hebben op mens en milieu. De schimmel Mortierella hyalina is een veelbelovend biologisch alternatief.

Chemische oorlogsvoering

Op de dag van vandaag worden goedkope pesticiden systematisch gebruikt als opbrengstverzekering, in plaats van sporadisch in het geval van een plaag. Dit leidt ertoe dat de consument, maar vooral ook de boer, sporen van pesticiden consumeert of inhaleert met mogelijk dramatische gevolgen voor de gezondheid. Vooral in derdewereldlanden, waar regelgeving rond het gebruik van pesticiden minder of zelfs niet aanwezig is, worden boeren in zeer grote mate blootgesteld aan kankerverwekkende chemicaliën die bovendien kunnen leiden tot problemen bij de zwangerschap en geboorteafwijkingen. Daarenboven zorgt overmatig gebruik van pesticiden voor de evolutie van zogenaamde “superpathogenen” die een resistentie opbouwen tegen deze chemische bestrijdingsmiddelen en alzo in de toekomst nog grotere plagen kunnen veroorzaken.

Ook kunstmest wordt systematisch gebruikt om de opbrengst te verhogen. Wanneer kunstmest toegediend wordt aan de bodem komen er in één keer zodanig veel nutriënten vrij dat het lang kan duren alvorens dat ze gefixeerd worden door de plantenwortels en bodemmicroben. Een ernstig gevolg hiervan is uitspoeling: het fenomeen dat zware regenval of irrigatie de voedingsstoffen en mineralen (vooral stikstof) uit de kunstmest wegspoelen waardoor het grondwater geëutrofieerd wordt. Eutrofiëring is nefast voor de biodiversiteit van naburige ecosystemen omdat snelgroeiende soorten (meestal algen) het gemakkelijkst gebruik kunnen maken van de overvloed aan stikstof.

U kan zien dat er dringend nood is aan duurzame alternatieven die de opbrengst en kwaliteit van land- en tuinbouwgewassen waarborgen, om het gebruik van gevaarlijke chemicaliën drastisch te verminderen.

Insecten, spinnen en schaaldieren

Chitine is alomtegenwoordig in de natuur. Schimmels, rondwormen, insecten, spinnen en schaaldieren bestaan grotendeels uit deze structurele molecule. Onderzoek heeft aangetoond dat het toevoegen van chitine aan de bodem gunstige gevolgen heeft voor bodemkwaliteit, plantengroei en het immuunsysteem van planten. Bovendien is er geen sprake van eutrofiëring, want het voedzame chitine kan maar langzaamaan afgebroken worden in het ecosysteem, waardoor er nooit een plotse overvloed aan stikstof voorkomt.

De schimmel Mortierella hyalina speelt hierin een cruciale rol.

Net zoals de darmflora bij mensen een grote invloed heeft op de gezondheid (“goede” en “slechte” bacteriën), zijn de microben rond de plantenwortels van fundamenteel belang voor een goede en gezonde ontwikkeling van de plant. Het microbioom rond het wortelstelsel is zelfs zo invloedrijk dat dit soms het tweede genoom van de plant wordt genoemd! Daarom onderzochten we hoe chitinetoediening aan de voedingsbodem van slaplantjes het microbioom van de slaplant beïnvloedt. De chitinebehandeling veroorzaakte een gigantische verandering in de schimmelgemeenschap die rond de wortels leeft. Eén schimmel stak er met kop en schouders bovenuit: Mortierella hyalina.

Maar hoe komt dat juist?

Het ABC van DNA

Waar je als organisme al dan niet toe in staat bent, zit gecodeerd in je genoom. Een genoom bestaat uit genen die coderen voor eiwitten, die op hun beurt nagenoeg alle functies in een biologisch lichaam uitvoeren. Beschouw eiwitten als kleine fabriekjes die bijvoorbeeld energie produceren, stikstof fixeren of chitine afbreken. Je genen zijn dan het bouwplan voor deze fabriekjes. De codetaal waarin die genen geschreven staan noemen we DNA. Je genoom is dus een boek van DNA dat voor ieder individu anders is. Bio-informaticamethoden laten toe om die cryptische maar levensnoodzakelijke informatie te ontcijferen.

Om een verklaring te vinden voor het immense succes van Mortierella hyalina in een chitinerijk milieu kan je dus een functionele genoomanalyse doen, waar je op zoek gaat naar genen die de waargenomen effecten veroorzaken, zoals genen die actief zijn in chitine-afbrekende systemen. Maar dat is niet alles. De bekomen DNA sequentie bevat immers alle informatie die Mortierella hyalina als organisme definieert. Daarom zijn we ook op zoek gegaan naar genen die instaan voor plantengroeibevordering en het versterken van het plantaardig afweersysteem.

De resultaten zijn verbluffend. Mortierella hyalina is in staat om chitine op twee verschillende manieren af te breken en om te zetten naar plantengroeibevorderende componenten (zie figuur). Enerzijds kan het met de eiwitten “N-acetyl-glucosaminidase” en “endochitinase” chitine omzetten naar N-acetyl-glucosamine, of GlcNAc in het kort. Anderzijds kan chitine ook geconverteerd worden naar chitosan, met het “chitine deacetylase” eiwit. In de literatuur is het een geweten feit dat GlcNAc en chitosan een plantengroeibevorderende werking vertonen. Bovendien vonden we genen die een rol spelen in stikstoffixatie, wat belangrijk is om uitspoeling te bestrijden, en genen die het afweersysteem van gewassen versterken.

Schimmel op je sla?

Wat betekent dit nu concreet? Wel, dit alles toont aan dat Mortierella hyalina veel potentieel heeft om een belangrijke speler te worden in duurzame gewasbescherming en -bebouwing. De biotechnologie is de laatste jaren erg sterk gevorderd en we zijn nu in staat om de goede aspecten van zulke bodemorganismen genetisch te versterken en eventuele negatieve aspecten te minimaliseren. Hoewel dit concept nog niet op industriële schaal ontwikkeld is, zijn de resultaten uit ons labo veelbelovend. Door potgrond of een akkerbodem te enten met een Mortierella hyalina stam, worden de planten op een natuurlijke manier sterker. Het toevoegen van chitine uit krabbenafval, of simpelweg insecten in de bodem, maakt dit effect nog veel intenser. Zo kunnen we een belangrijke stap zetten naar een toekomst zonder pesticiden en chemische meststoffen.

En maak je geen zorgen, de sla die je in de winkel koopt zal niet beschimmeld zijn. Mortierella leeft in de bodem, en bovendien vindt deze schimmel sla niet eens zo lekker. Dode insecten daarentegen? Jammie!
