Toxin variation in Alpine salamander populations

“Mijn gif is het jouwe niet”. Hoe het gif van salamanders wordt beïnvloed door de omgeving.

De Romeinse auteur Plinius de Oudere schreef ooit dat de salamander “het meest valse van alle giftige dieren” was en wijdde uit over hoe deze dieren hele dorpen konden vergiftigen. Ook in de middeleeuwen werden salamanders, kikkers en padden vaak geassocieerd met zwarte magie omwille van hun giftige aard. Ondertussen heeft de afkeer voor deze dieren plaats gemaakt voor fascinatie. Doorheen de jaren is de samenstelling van amfibieën-gif dan ook uitgebreid bestudeerd, zowel uit wetenschappelijke nieuwsgierigheid als met het oog op medische toepassingen. Opmerkelijk is echter dat zelfs binnen één soort, amfibieën enorm kunnen verschillen in de samenstelling van hun gif, en dus de mate waarin ze giftig zijn.

Over leven en gif

 “Het zijn niet de sterkste soorten die overleven,” schreef Charles Darwin, de grondlegger van de evolutietheorie ooit, “en ook niet de meest intelligente. Het is de soort die het beste reageert op veranderingen.” Amfibieën vormen hier geen uitzondering op. Als een salamander of kikker wil voorkomen dat hij opgegeten wordt, moet het méér dan alleen giftig zijn. Zijn gif moet zodanig samengesteld zijn, dat het hem optimaal beschermt tegen de natuurlijke vijanden in zijn specifieke omgeving. Deze natuurlijke vijanden zijn niet alleen vogels of slangen die graag amfibieën lusten, maar eveneens micro-organismen, zoals bacteriën en schimmels, die de kwetsbare en vochtige huid van deze dieren zouden kunnen infecteren. In een omgeving waar veel schimmels aanwezig zijn, zou het gif van een salamander of kikker dan ook in principe voldoende stoffen moeten bevatten die de groei van schimmels tegen gaan. In een omgeving waar veel roofdieren aanwezig zijn, zou een amfibie voldoende stoffen moeten produceren die vijanden afschrikken, verlammen of zelfs doden.

Maar waarom zou een salamander, kikker of pad niet gewoon zo giftig mogelijk zijn als maar kan? Wat houdt amfibieën tegen om een karrevracht aan gifstoffen te produceren die hen beschermen tegen zoveel mogelijk natuurlijke vijanden? Dit is voornamelijk te wijten aan het feit dat in de natuur alles een kost heeft. De aanmaak van gifstoffen is energetisch duur, en alle energie die wordt geïnvesteerd in gif, kan niet meer worden geïnvesteerd in andere zaken, zoals voortplanting en groei. Dit wordt een trade-off genoemd. In een omgeving waar het risico om geconsumeerd te worden laag is, loont het niet om te investeren in een overdaad aan gifstoffen. Om deze reden zou men verwachten dat de samenstelling van gif afhankelijk is van het risico op predatie en infectie in de omgeving.

Salamanders in de bergen

Hoe test men echter zoiets? Ten eerste had ik een studiesoort nodig die giftig was, maar ook voorkomt in diverse omgevingen die verschillen in het risico op predatie en infectie. De keuze viel op de enigmatische alpensalamander, een familielid van onze vuursalamander. Deze salamander kan gevonden worden van dichtbegroeide loofbossen op gemiddelde hoogtes tot bergtoppen waar men slechts rotsblokken en gras vindt. Uiteraard vereiste dit het nodige klimwerk om de salamanders te kunnen vangen, wegen, meten en hun gif te kunnen verzamelen. Het verzamelen gebeurde in Kroatië, Montenegro en Bosnië en Herzegovina. Het gif zelf werd geanalyseerd in België. Specifiek werd gekeken naar twee componenten: samandarine, dat volgens eerdere experimenten het meest krachtige bestanddeel van dit gif is, en samandarone, dat een duidelijk negatief effect heeft op de groei van schimmels en bacteriën.

Ten tweede is het noodzakelijk om een idee te krijgen van hoe groot het risico op infectie is in elke omgeving. Aangezien de alpensalamander het grootste deel van zijn leven onder de grond doorbrengt, werden van elke locatie bodemstalen meegenomen. Aan de hand van moleculaire technieken kon zo achterhaald worden hoeveel en welke bacteriën en schimmels er overal voorkwamen. Verder moet men ook weten hoeveel dieren er in de omgeving aanwezig zijn die potentieel deze salamanders kunnen opeten. Dit was waarschijnlijk het minst opwindende deel van mijn proefwerk, aangezien dit voornamelijk gebeurde op basis van literatuurgegevens en online databases.

De uitkomst

Het samenbrengen van deze gegevens leverde een mix op van verwachte en onverwachte resultaten. Daar waar salamanders veel predatoren hebben, bevat hun gif vreemd genoeg de kleinste hoeveelheden van samandarine, terwijl salamanders die leven in een omgeving waar nauwelijks predatoren voorkomen, juist relatief veel samandarine produceren. Interessant genoeg bleek wel dat de grootste salamanders werden gevonden daar waar de meeste soorten slangen voorkwamen, zeker aangezien grotere salamanders meer gif kunnen opslaan in hun lichaam en dus giftiger zijn. Eveneens was het zo dat salamanders in omgevingen met meer bacteriën en schimmels in de bodem zoals verwacht meer samandarone afscheiden.

Er zijn verschillende redenen waarom de resultaten de gestelde hypothesen slechts gedeeltelijk bevestigen, maar wellicht de belangrijkste is dat er veel meer factoren een rol spelen bij gif-variatie. Mijn thesis vormt dan ook een interessant vertrekpunt voor verder onderzoek, waarbij stap voor stap de redenen voor gif-variatie kunnen ontmaskerd worden.

Tot slot, wat heeft dit voor een nut, afgezien van wetenschappelijke interesse? De laatste jaren worden talloze amfibieën-soorten bedreigd door de schimmelziekte chytridiomycosis. In het zuiden van Nederland is het merendeel van de vuursalamanders aan deze ziekte is bezweken. Maar niet alle amfibieën blijken even vatbaar te zijn voor deze ziekte, en de samenstelling van hun gif zou hier een belangrijke rol in kunnen spelen. Het bestuderen van gif-variatie kan dan ook een belangrijk hulpmiddel worden om in te schatten welke soorten en welke gebieden extra kwetsbaar zijn. Tenslotte zou het zonde zijn om kikkers, padden en salamanders van de aardbodem te zien verdwijnen, ongeacht hoe “slecht en vals” Plinius de Oudere hen ook vond.
