Effecten van twee additieven ‘selenium en cysteïne’ op hittestress bij vleeskippen.

Tijdens de zomermaanden kunnen hoge buitentemperaturen zorgen voor een verhoogde stressgevoeligheid bij vleeskippen. Er wordt gesproken van hittestress als de warmteafvoer onvoldoende groot is, waardoor het lichaam van de kippen opwarmt. Zo’n stressfactor zorgt in de pluimveeteelt voor verminderde voederopname en een daaropvolgende mindere groei. Daaruit kan uiteraard een slechtere voederconversie (>1,80) optreden. Om dit probleem aan te pakken, werd in het kader van mijn masterproef het effect van selenium en het aminozuur cysteïne onderzocht. Deze stoffen zouden bepaalde antioxidanten verhogen, die op hun beurt de vorming van schadelijke radicalen kunnen verminderen, waardoor de stress in het lichaam daalt.

Selenium werd toegediend onder de vorm van selenium-methionine en cysteïne onder de vorm van N-acetyl-cysteïne, aangezien deze stoffen stabieler zijn. Het effect van deze stoffen op het lichaam werd onderzocht door middel van weefselonderzoek.

De proef werd uitgevoerd in het proefcentrum Lanupro van de Universiteit Gent te Melle. Er werden 680 vleeskippen verdeeld over 34 hokken en er werden acht groepen gemaakt. Alle kippen kregen fase voedering. Eén groep werd ter beschikking gesteld als controlegroep en kreeg bijgevolg geen selenium en cysteïne toegediend. De zeven overige groepen kregen ofwel enkel selenium, ofwel enkel cysteïne ofwel kregen ze beide additieven in het voeder. Cysteïne werd toegediend in verschillende hoeveelheden. De dieren werden van dichtbij opgevolgd. Bij elke faseovergang werd de hoeveelheid resterende voeder gewogen, zodat we konden nagaan hoeveel de kippen in één fase hadden gegeten. Ook werden de dieren bij iedere faseovergang gewogen. Selenium werd aan het voeder toegevoegd vanaf dag 1, terwijl cysteïne pas werd toegevoegd vanaf dag 25. Vanaf dag 28 werd het hittestressmodel toegepast. Vanaf deze dag werden de temperaturen verhoogd tot 34°C om zo de warme zomermaanden na te bootsen. ’s Nachts daalde de temperatuur terug naar 22°C.

De proef duurde zoals in de realiteit 42 dagen. De effecten van de additieven werden onderzocht op acuut niveau en op chronisch niveau. Dit wil zeggen dat enerzijds de dag na de hogere temperaturen de kippen werden geslacht en onderzocht en anderzijds de kippen werden geslacht na een lange periode van warme temperaturen. Per keer werden 68 kippen geslacht.

Wat gebeurt er in het lichaam bij hittestress?

Door overtollige warmte in het lichaam van de kippen kunnen radicalen ontstaan, omdat de processen ter hoogte van de mitochondriën minder efficiënt werken. Deze radicalen zijn schadelijk voor het lichaam en kunnen zorgen voor verzwakking van de dieren, mindere eetlust, minder goede processen in het lichaam. De perioden van hittestress kunnen zorgen voor heel wat sterfte bij de dieren. Door het toevoegen van deze additieven worden de radicalen in aantal verminderd en blijven de dieren langer gezond.

 

Resultaten van de proef:

  • Rectale temperatuur

Door het meten van de rectale temperatuur kon geconcludeerd worden dat het zeker om hittestress ging. In normale omstandigheden bedraagt de lichaamstemperatuur van een vleeskuiken 41°C. In omstandigheden van overvloedige warmte kan deze temperatuur snel oplopen tot 42,5 tot 43°C.

  • Groeiprestaties:

Bij toevoeging van N-acetyl-cysteïne die het belangrijk aminozuur cysteïne bevat, bleek er een verhoogde groei te zijn. De kippen groeiden in het algemeen wel minder vlug dan in een situatie zonder hittestress. Toch werden er snellere groeiprestaties waargenomen bij de vleeskippen die cysteïne toegediend kregen in vergelijking met de controlegroep. Er zijn resultaten gemeten van 2844 gram op het einde van de groeiperiode (dag 41). Zonder toevoeging van cysteïne was dit slechts 2675 gram. Het normale gewicht volgens ROSS 308 bedraagt 2917 gram, waardoor besloten kan worden dat de kippen wel onder het gemiddelde bleven. Cysteïne had niet alleen een positief effect op de groei, maar er werd ook een betere voederopname vastgesteld. Bij toevoeging van cysteïne, verkregen de dieren met mindere opname van voeder toch een hoger gewicht. Dit kan ook waargenomen worden in een veel verbeterde voederconversie. Deze daalde van 2.21 naar 1.8 bij enerzijds geen en anderzijds wel toevoeging van cysteïne.

Ook selenium had een positief effect op de groei. Bij toevoeging van 0,20 mg/kg Se werd een verhoogde groei waargenomen op dag 10. Ook de dagelijkse aanwinst was beter bij toevoeging van selenium. Echter op latere leeftijd kon selenium geen verschil meer maken, waardoor we kunnen besluiten dat selenium best wordt toegevoegd in de startersfase en cysteïne beter wordt toegevoegd in de afmestfase. Alle resultaten zijn weergegeven in de volgende tabellen.

  • Fysiologische parameters

De effecten van selenium en cysteïne op het antioxidant glutathion werden ook nagegaan. Glutathion is een antioxidant die zorgt voor een vermindering van het aantal radicalen, waardoor de stress in het lichaam van de kippen daalt. In dit onderzoek zijn er geen opmerkelijke verschillen gevonden. Er kon wel aangetoond worden dat bij acute hittestress de lever veel meer oxidatieve stress vertoont, wat wil zeggen dat in de beginfase van de warme temperaturen de lever het meeste stress ondergaat. In het begin van een warme periode is het dus aan de lever om te zorgen voor de productie van antioxidanten. Deze worden vervoerd in het bloed en zorgen in heel het lichaam voor verminderde vrijgave van radicalen. Dit kon tevens verklaard worden uit resultaten van het hartweefsel. Het hart heeft weinig invloed van oxidatieve stress in de beginfase, aangezien de antioxidanten uit de lever de radicalen verminderen. Het hart blijft onder controle door de lever, waardoor minder sterfte optreedt.

In deze masterproef kon tevens geconstateerd worden dat na chronische hittestress de antioxidanten in de lever daalden. Dit omdat er in de andere organen teveel vraag is naar antioxidanten, waardoor de lever onvoldoende kan aanmaken om aan deze vraag te voldoen. Het is dus met andere woorden belangrijk in het begin van de opkomende hitte snel in te grijpen.

  • Effect van cysteïne op aminozuur methionine

Door het toedienen van het aminozuur cysteïne kan een sparend effect verkregen worden van een andere heel belangrijk aminozuur: methionine. In het lichaam moet methionine eerst worden omgezet in cysteïne, pas dan kan het gebruikt worden voor de aanmaak van dat antioxidant. Deze omzettingsprocessen vragen uiteraard ook weer energie, waardoor er minder over is voor andere processen, zoals de groei. Daarom dat cysteïne op dit vlak efficiënter is tegen hittestress dan methionine. Door toevoeging van cysteïne wordt op deze manier ook methionine gespaard, waardoor dit aminozuur beter kan gebruikt worden in andere processen en ook hier weer kan dit beter zijn voor de groei.
