HEK293 GlycoDelete: optimalisatie door vergelijking van verschillende endoglycosidasen

De kap van suikerbomen gaat onverbiddelijk door!

“Ik heb nog nooit een suikerboom gezien, in welk sprookjesland komen deze voor?” hoor ik u al vragen. Deze bomen bezetten het oppervlak van eiwitten, dus niet meteen vergelijkbaar met een doordeweekse berk of beuk. Recent werd een technologie ontwikkeld om deze suikerbomen weg te maaien. Een vorm van ontbossing met vele voordelen, vooral in de farmaceutische industrie.

Eiwitten zijn één van de voornaamste bouwstenen van ons lichaam, ze komen voor in en op elke cel en stromen rijkelijk in het bloed. Vroeger werd reeds gedacht dat eiwitten als medicatie zouden kunnen gebruikt worden. In de laatste jaren is dit ook werkelijkheid geworden met als resultaat dat er vandaag de dag meer dan 200 therapeutische eiwitten op de markt zijn. Eén belangrijk voorbeeld is ‘Etanercept’, een eiwit voor behandeling van reuma.

Deze therapeutische eiwitten hebben duidelijk aan belang gewonnen, maar de productie ervan is echter nog steeds een uitdaging. De biofarmaceutische industrie gebruikt hiervoor vooral zoogdiercellijnen, dus ook menselijke cellijnen. Deze cellen decoreren het oppervlak van de geproduceerde eiwitten met zeer variabele suikerbomen. Daarenboven wordt de vorm van deze suikerstructuren extra beïnvloed door parameters waaronder de cellen worden gegroeid, zoals zuurtegraad en temperatuur. Het geproduceerde therapeuticum is dus een mengsel van eenzelfde eiwit met suikerbomen die verschillen in het aantal vertakkingen. De dienst Volksgezondheid eist dat het product steeds dezelfde kwaliteit heeft, maar de variabiliteit van suikerbomen maakt dit niet eenvoudig.

Ernstiger is wanneer een biofarmaceuticum, zoals een vaccin, zijn therapeutische werking verliest omwille van de aangehechte suikerbomen. Elk jaar moet u een nieuw griepvaccin krijgen omdat hetgeen dat u vorig jaar kreeg geen bescherming meer biedt. Hoe kan dat nu? Influenza is één van die snel veranderende virussen. Het virus trekt als het ware elk jaar een ander jasje aan. Deze mantel bestaat uit eiwitten. Op deze eiwitten zijn wel patronen die minder snel veranderen. Vaccins die een immuunrespons opwekken tegen deze patronen zouden een langdurige bescherming kunnen bieden tegen het virus. Het virus heeft echter die meer geconserveerde patronen bomvol suikerbomen gezet. Deze suikerbomen fungeren als een soort paraplu waaronder de patronen verscholen worden voor ons immuunstelsel.

Als deze suikerbomen zoveel problemen veroorzaken, waarom maaien we deze dan niet gewoon weg? Suikerbomen hebben een belangrijke functie in het vouwen van de eiwitten tot de juiste vorm en kunnen niet zomaar verwijderd worden. Na correcte opvouwing hebben de meeste suikerstructuren slechts een beperkte functie. Dus nadat ze hun cruciale rol in eiwitopvouwing gespeeld hebben zouden ze eventueel verwijderd kunnen worden.

Recent werd de GlycoDelete-technologie ontwikkeld die exact dit doet. Bij deze technologie werd in een menselijke cellijn via genetische manipulatie een soort kettingzaag (een endoglycosidase) ingebouwd. Het endoglycosidase kapt suikerbomen om die op het oppervlak van eiwitten zitten. Maar dit endoglycosidase vereist dat deze suikerbomen allemaal van dezelfde vorm zijn. Dit wordt verkregen door extra genetische modificatie van de menselijke cellijn. Concreet werd een gen dat instaat voor het veranderen van de vorm van de suikerboom verwijderd.

Vooral de farmaceutische industrie toont interesse in deze technologie, maar de GlycoDelete-technologie is tot op heden nog niet zo makkelijk in gebruik en moest daarom geoptimaliseerd worden. Vooral de extra modificatie om alle suikerbomen van dezelfde vorm te maken, is niet goedkoop en niet eenvoudig. In deze thesis werden verschillende soorten endoglycosidasen getest. Een bepaalde variant van endoglycosidasen bleek bovenverwachte activiteit te vertonen. Dit endoglycosidase kon alle vormen van suikerbomen verwijderen. Het endoglycosidase werd verder gekarakteriseerd en tot dusver zijn de eerste resultaten veelbelovend.

Er werd dus een ‘shortcut’ gevonden in de GlycoDelete-technologie die deze makkelijker, sneller en goedkoper kan maken. Dit kan het duwtje in de rug zijn zodat de technologie ingang vindt in de farmaceutische industrie, waardoor goedkopere en efficiëntere medicatie kan gevonden worden.
