Effects of copepods, diatoms and their interactions on denitrification in marine sediments: an experimental approach

In de Westerschelde worden dagelijks grote hoeveelheden nitraat geloosd. Wanneer deze stof in zee stroomt, wordt het natuurlijk biochemisch evenwicht verstoord. Gelukkig zijn er bacteriën in de riviermonding aanwezig, die het nitraat kunnen omzetten tot onschadelijk stikstofgas. Willem Stock onderzocht in zijn masterproef hoe deze micro-organismen beïnvloed worden door de organismen waarmee ze samenleven.

Alle levende organismen hebben stikstof nodig. Het is een essentieel onderdeel van eiwitten en DNA. Stikstof maakt zo’n 78% uit van de atmosfeer, doch niet altijd in de gepaste vorm die opname door planten en dieren toelaat. De meeste organismen zijn immers niet in staat om stikstofgas op te nemen. Slechts enkele bacteriën kunnen stikstofgas opnemen en omzetten naar actief stikstof, zoals ammonium en nitraat, waar de andere organismen wel gebruik van kunnen maken.

In de lente van 1914, in het kleine Duitse stadje Oppau, werd de eerste fabriek in gebruik genomen die op grote schaal ammonium kon produceren uit stikstofgas. De opening van de fabriek luidde het begin in van de meest drastische impact op de globale biochemie ooit. Nu, net geen honderd jaar later, is door de menselijke activiteit de hoeveelheid actief stikstof op Aarde verdubbeld. In vele Europese riviermondingen is de actieve stikstoftoevoer zelfs vertienvoudigd. Door de hoge stikstofbelasting ontstaan giftige algenboeien en wordt de samenstelling van de aanwezige fauna en flora sterk verstoord. Om de gevolgen van de verhoogde stikstofbelasting te kunnen remediëren, is het in de eerste plaats noodzakelijk dat we de stikstofcyclus en de rol van aquatische organismen hierin beter begrijpen.

In het slib van riviermondingen zijn bacteriën aanwezig die in staat zijn om actief stikstof te verwijderen. Ze zetten de overmaat aan nitraat en nitriet terug om tot stikstofgas tijdens een proces genaamd denitrificatie. Via dissimilatieve nitraatreductie, zijn er zelfs bacteriën die nitraat en nitriet kunnen omzetten naar ammonium. Ammonium zal, in tegenstelling tot de gassen gevormd door denitrificatie, niet verdampen. In tegendeel, ammonium is voor veel organismen, zoals algen, makkelijker op te nemen dan nitraat.

Beide processen gebeuren simultaan, naast elkaar. De bacteriën, die de reacties katalyseren, treden er in competitie voor het beschikbare nitraat en nitriet. Welke bacteriën er de overhand hebben, wordt bepaald door de omgevingscondities. Met andere woorden, het zijn deze condities die bepalen of de actieve stikstof in het systeem blijft of dat ze eruit verwijderd wordt. Als we de zware stikstofbelasting in onze riviermondingen en kustzones willen tegen gaan, is het bijgevolg noodzakelijk om te weten welke spelers die condities beïnvloeden.

In zijn scriptie focuste Willem Stock zich op de invloed die kiezelwieren en roeipootkreeftjes hebben op de condities in het slib van de Westerscheldemonding en de gevolgen hiervan op denitrificatie en dissimilatieve nitraatreductie. Kiezenwieren zijn eencellige algen die vaak zeer talrijk bovenop het slib aanwezig zijn en vormen er een donkerbruine, zogenaamde biofilm die duidelijk zichtbaar is op het slib. De biofilm vormt de basis van het voedselweb in deze sedimenten. Roeipootkreeftjes zijn kleine kreeftachtigen en belangrijke grazers van zowel kiezelwieren als bacteriën. Zowel kiezelwieren als roeipootkreeftjes komen ze in grote aantallen voor in het slib, waar ook bacteriën aanwezig zijn die nitraat en nitriet gebruiken.

Om het effect van de algen en de kreeftjes op de stikstoffluxen te onderzoeken werden kleine experimentele containers ingericht. Slib, verzameld in de Westerscheldemonding, werd gelijk verdeeld over de containers. Het slib werd vooraf gezeefd en ingevroren om er alle algen en roeipootkreeftjes uit te verwijderen. De bacteriën zijn in staat het invriezen te overleven en zij bleven er dus in aanwezig. Aan de containers werden vervolgens kiezelwieren, roeipootkreeftjes, een combinatie van beiden of niets toegevoegd. Na een week incubatie onder gecontroleerde licht- en temperatuurcondities, werd een grote hoeveelheid nitraat toegevoegd. Door de concentratieveranderingen van nitraat en de andere stikstofcomponenten op te volgen, kon bepaald worden welke invloed de algen of kreeftjes en hun interacties hadden op de nitraat-omzettende bacteriën. De roeipootkreeftjes bleken een significante invloed te hebben. In hun aanwezigheid werd minder nitraat omgezet via denitrificatie ten voordele van dissimilatieve nitraatreductie. Er werd dus meer actief stikstof in het systeem gehouden in hun aanwezigheid. Dit geobserveerde effect was hoogst waarschijnlijk te wijten aan de excretieproducten van de kreeftjes. Ze vormen een belangrijke bron van koolstof voor de bacteriën en vooral diegene die nitraat tot ammonium omzetten weten hier goed mee om te gaan. De kiezelwieren op zich bleken geen significante invloed te hebben. In de containers waar zowel algen als kreeftjes aanwezig waren, was de invloed van de roeipootkreeftjes nog sterker. De kiezelwieren konden dus, hoewel ze zelf geen effect hadden op de stikstoffluxen, wel het effect van de kreeftjes versterken. Ze waren immers een extra voedselbron voor de roeipootkreeftjes, waardoor deze meer excretieproducten uitscheidden.

Het is duidelijk een strategie van de roeipootkreeftjes om de actieve stikstof in het slib te houden. Dit actieve stikstof zal immers de groei van kiezelwieren en bacteriën bevorderen. De roeipootkreeftjes zorgen op die manier voor een versterkte groei van hun voornaamste voedselbronnen.

Aan de hand van labo-experimenten werd in deze masterproef voor de eerste maal aangetoond dat roeipootkreeftjes een belangrijke impact kunnen hebben op de hoeveelheid actief stikstof die uit de riviermonding verwijderd wordt. Dit is een essentiële bijdrage tot onze, tot nog toe, beperkte kennis van de stikstofcyclus en de factoren die de stikstofconcentraties in aquatische milieus bepalen. Het is een belangrijke stap voorwaarts in het inschatten van de gevolgen die de grote input van actieve stikstof door antropogene activiteit heeft en zal hebben in de toekomst.
