Confocale microscopietechnieken in het onderzoek naar het werkingsmechanisme van een nieuw en uniek anti-HIV middel

Alvorens HIV de gastheercel binnendringt, bindt het virale gp120 aan de CD4-receptor van de gastheercel. Eenmaal het HIV gebonden is ondergaat het gp120 een conformatieverandering wat uiteindelijk leidt tot het versmelten van de membranen van de gastheercel en het virus. Het CADA-product is in staat de CD4-receptor te downmoduleren. Dit zorgt ervoor dat er geen binding meer zal zijn tussen het gp120 van HIV en de CD4-receptor van de gastheercel. HIV kan niet meer binnendringen in de gastheercellen, niet meer repliceren en dus ook geen nieuwe virions meer vormen.

 

Met behulp van flowcytometrie en verschillende monoclonale antilichamen werd het werkingsmechanisme van het antivirale CADA-product in detail onderzocht. Nadat Hela CD4+ cellen en MT4 (CD4+) cellen behandeld werden met CADA werd er een duidelijke CD4-downmodulatie vastgesteld. De procentuele CD4-downmodulatie was voor beide cellijnen identiek. De CD4-downmodulatie is dus onafhankelijk van het type cellijn. Door gebruikt te maken van de competitie tussen verschillende mAb voor bindingsplaatsen op de CD4-receptor, is gebleken dat het CADA-product niet bindt op het extracellulaire gedeelte van de de CD4-receptor. Na behandeling van de Hela CD4+ cellen met CADA en incubatie met verschillende mAb, die elk binden op een andere bindingsplaats op de CD4-receptor, werd een identieke procentuele verlaging van het emissiesignaal vastgesteld. De behandeling van CD4+ cellen met CADA resulteert in een downmodulatie van de volledige CD4-receptor.

 

Om het werkingsmechanisme van het CADA-product beter te begrijpen, werd er gewerkt met een fluorescente analoog nl. KKD-016. Uit voorafgaand onderzoek is gebleken dat KKD-016 dezelfde antivirale en biologische eigenschappen heeft als CADA. Met behulp van confocale microscopie kunnen we het KKD-016 zien in de cellen omwille van zijn fluorescente eigenschap. Het product wordt in de cellen opgenomen afhankelijk van de aanwezige concentratie van het product in het cultuurmedium. Deze dosisafhankelijke opname toont flowcytometrisch een dosisafhankelijke effect op de CD4-downmodulatie. Zelfs na 2 uur incubatie van de Hela CD4+ cellen met KKD-016 is er een duidelijke opname van het product. Verder werd met behulp van kern- en membraankleuringen vastgesteld dat het product (KKD-016) niet doordringt tot in de kern van de cellen maar zich lokaliseert in het cytoplasma.

 

Door Hela cellen te transfecteren met CD4-YFP brengen deze cellen een fluorescente CD4-receptor tot expressie. Met het oog op het bekomen van een sterk CD4+ cellijn werden de CD4+-getransfecteerde cellen geïsoleerd uit het celmengsel en verder opgekweekt.

Wanneer deze cellen na het opkweken in dezelfde mate de fluorescente CD4-receptor tot expressie brengen, kunnen ze worden behandeld met CADA of KKD-016 om de CD4-downmodulatie verder te onderzoeken.
