The effects of chronic oxytocin treatment in a mouse model of delayed brain development

Oxytocine en sociaal gedrag: it’s all in the details.

De term ‘oxytocine’ mag dan wel exotisch klinken, als mens staan we continu onder zijn invloed. Bij wie nog steeds geen belletje is gaan rinkelen, zullen termen als ‘knuffelhormoon’ of ‘liefdeshormoon’ zeker helpen. Toch doen deze laatste twee termen sterk afbreuk aan de invloed die oxytocine heeft op ons gedrag. Na jarenlang onderzoek lijkt dit hormoon en zijn functie voornamelijk te zijn gehuld in tot op het heden onverklaarbare raadsels.

Ergens tussen hoop en hype

In 1970 wezen een reeks onderzoeken uit dat het hormoon oxytocine een rol speelt in enerzijds de band tussen ouder-kind en anderzijds de band tussen (romantische) partners. Dit zette zowel de wetenschappelijke wereld alsook de commerciële wereld op zijn kop. Het veelbelovende idee ontstond dat een spray oxytocine iemand aantrekkelijker of zelfs meer zelfzeker kan maken. De wetenschappelijke wereld was niet minder ambitieus. Hier werd de claim geuit dat een spray oxytocine in de neus de sociale effecten van psychiatrische stoornissen, gaande van autisme spectrum stoornissen tot verslaving, kon verminderen of zelfs verhelpen.

De wafel-analogie

In werkelijkheid blijkt de werking van het hormoon meer complex te zijn. Laat ons het probleem tastbaar maken door een analogie: grootmoeders wafels. De wafels van mijn grootmoeder zijn ongetwijfeld de lekkerste, al zal u waarschijnlijk hetzelfde denken. Het vreemde is dat de basis-ingrediënten voor wafels altijd dezelfde zijn: boter, melk, bloem,… De kunst echter zit hem in een aantal aspecten: in welke hoeveelheid gebruik ik de ingrediënten voor een goede balans? Meng ik  het deeg best handmatig of met een mixer? Wanneer doe ik het deeg in het wafelijzer? Hoe lang moet het deeg in het wafelijzer?

Het principe is hetzelfde bij oxytocine. In de eerste plaats maakt het een verschil uit in welke hoeveelheid deze stof wordt toegediend. Onderzoek heeft vele verschillende hoeveelheden gebruikt.
Toch zijn wetenschappers na 50 jaar intensief onderzoek het nog steeds niet eens over de optimale hoeveelheid om toe te dienen. Ten tweede was er het praktische aspect: hoe meng je het perfecte deeg? In geval van oxytocine wordt dit best vertaald naar hoe we het hormoon toedienen. Er zijn verschillende mogelijkheden. Men kan het innemen in de vorm van een tablet, inspuiten in de bloedstroom of in de hersenen. Momenteel is de meest gebruikte wijze echter het toedienen in de vorm van een neusspray.

Vervolgens hangt ons resultaat van de oxytocine op sociaal gedrag af van de leeftijd van de persoon. Dan is er ook nog de vraag hoe lang we het hormoon zullen toedienen. De meeste onderzoeksresultaten die socialer gedrag rapporteren, dienen de oxytocine eenmaal toe. Vanuit wetenschappelijke eerlijkheid moet dan echter de nuance gemaakt worden dat psychiatrische stoornissen meestal levenslang zijn. Dat betekent dat deze individuen niet geholpen zijn met een eenmalige toediening. Een laatste en waarschijnlijk belangrijkste aspect om de complexiteit te begrijpen is de omgeving. Bij het bakken van wafels gaat het dan om het samenspel tussen de verschillende ingrediënten: vanille gaat bijvoorbeeld harmonieus samen met boter. Op een zelfde wijze dient er ook harmonie te zijn tussen oxytocine en andere hormoonsystemen, zoals testosteron, of neurotransmittersystemen, zoals dopamine.

 

 

Van muis naar mens

Veel van wat we vandaag over onze (sociale) hersenen weten is gebaseerd op bevindingen uit knaagdieren. U zal zich dan misschien de vraag stellen: een mens is toch geen knaagdier? Een absoluut terechte opmerking. Er zijn veel verschillen tussen de hersenen van  muis en mens om maar te starten met bijvoorbeeld de grootte. Toch zijn er gelijkenissen die deze vergelijking toelaten. Niet alleen zijn wij en de knaagdieren zoogdieren, ook onze hersenstructuren zijn globaal gezien erg gelijkend. Zeker op vlak van sociaal gedrag. Het bestuderen van muizen heeft ons veel geleerd over wat er in de hersenen gebeurt wanneer wij ons aan iemand ‘binden’, zoals een ouder of een partner. Dat maakt de cirkel volledig, want dan zijn we weer in het speelveld van oxytocine gekomen.

Om het sociale gedrag en afwijkingen ervan naar mensen te vertalen, worden specifieke muismodellen gebruikt. Dit zijn muizen die ofwel een volledige stoornis ofwel een deel ervan afbeelden. Bijvoorbeeld: om een goed muismodel voor autismespectrumstoornissen te zijn zal aan bepaalde voorwaarden voldaan moeten worden. Dit doet men aan de hand van gedragstesten. Door het gedrag van de muis op deze manier te meten, kan men iets zeggen over de onderliggende genen of hersenstructuren die een rol spelen in de stoornis.

Concreet

We wilden onderzoeken welk effect oxytocine heeft op sociaal gedrag. Daarvoor hebben we niet enkel ‘normale’ muizen gebruikt, maar ook een muismodel voor vertraagde hersenontwikkeling. Doordat muizen erg sociale dieren zijn, bestaan er vele verschillende testen om dit te meten. Wij hebben vier proeven gebruikt die de muizen moesten doen. Zonder oxytocine bleek dat er een verschil was tussen normale muizen en het muismodel op gebied van sociaal gedrag. Waar de normale muis geneigd is sociale contacten aan te gaan leek het muismodel eerder sociale contacten te vermijden. Interessant genoeg vonden we dat, met een heel lage dosis oxytocine in de neus, het gedrag van het muismodel socialer werd. Meer nog, we vonden dat dit effect waarschijnlijk gevonden werd doordat oxytocine stress verlaagt. Kortom, doordat de sociale ‘angst’ lager werd gedroegen de muizen zich anders.

Echter, onze bevindingen waren niet allemaal rozengeur en maneschijn. We behandelden de muizen 30 dagen lang met oxytocine. Daarna waren we nieuwsgierig of er andere veranderingen waren dan het sociaal gedrag. Daarom lieten we de muizen ook een leertest doen. Hieruit bleek dat de muizen niet langer in staat waren om te leren na een intensieve training.

Het feit dat oxytocine een effect kan hebben op hoe goed muizen kunnen leren, wijst op het belang van testen op muizen. Momenteel staat de wetenschap nog niet zo ver dat oxytocine als een wondermiddel beschouwd kan worden. Er moet nog veel onderzoek gebeuren en andere mogelijkheden zullen nog ontdekt moeten worden. We zijn er nog niet, we hebben nog een lange weg te gaan. Toch ziet de weg van oxytocine er nog steeds veelbelovend uit.
