Plantenafval als waardevolle grondstof?

In tijden van de opwarming van de aarde en steeds afnemende voorraden aan fossiele grondstoffen, registreerde het “renewables global status report” van 2016 toch een stijging in verbruik van petroleum van 127 miljoen ton, 2,6 keer hoger dan de stijging in verbruik van hernieuwbare bronnen. Plantenafval biedt met cellulose echter waardevolle alternatieven voor petroleum gebaseerde producten. Cellulose is als het meest voorkomende bio-polymeer ter wereld een interessante potentiële leverancier van een waaier aan waardevolle producten.

Rendabiliteit van een bio-gebaseerde industrie

Door een steeds toenemend verbruik, komen de natuurlijke voorraden aan fossiele grondstoffen steeds meer onder druk te staan. Samen met het zich manifesteren van de gevolgen van de opwarming van de aarde, zorgt dit zo voor een steeds groeiende interesse in hernieuwbare grondstoffen en een evolutie naar een meer duurzame bio-gebaseerde industrie. Daar de huidige technologieën voor een duurzamere industrie hoofdzakelijk gebaseerd zijn op het gebruik van voedingsgewassen zoals bio-ethanol uit graan, zou grootschalig gebruik bijgevolg resulteren in stijgende voedselprijzen.

Een potentieel interessante alternatief is het gebruik van cellulose uit plantaardige reststromen zoals papierafval, pulp, vlasresten, (snoei-)hout, bermgras en landbouwresidu's. Aangezien cellulose het meest voorkomende bio-polymeer ter wereld is en de plantaardige materialen bij hun aanmaak reeds bijdroegen tot het CO2 verbruik, kunnen zij gezien worden als een duurzame hernieuwbare voorziening. Door daarbij vooral te focussen op reststromen, kan hierdoor ook de invloed op de grondstofprijzen voor andere industrieën beperkt worden.

In deze evolutie naar een duurzame economie, wordt de toepassing van cellulose voor de bio-ethanolproductie echter bemoeilijkt door zijn kristalliene structuur. Mede door deze structuur ontstaat namelijk een enorme weerstand tegenover conventionele enzymatische afbraak. Hoewel dit voordelig is voor de stevigheid van de plantencelwand, resulteert het in een nood aan een complex enzymatisch mengsel bij de voorbehandeling van cellulose tot hernieuwbare grondstof. Door het vaak ontoereikend afbraakvermogen van de klassieke enzymenmengsels, werd de rendabiliteit van cellulose als duurzaam alternatief voor fossiele grondstoffen vaak in twijfel getrokken.

De oplossing?

De recent ontdekte LPMO enzymen (Lytische Polysaccharide Mono-Oxygenases)  worden nu echter door vele onderzoeksgroepen aanzien als de ontbrekende factor voor een efficiënte enzymatische cellulose afbraak. Door hun opbouw kunnen ze zelfs in de kristalliene structuur de glucoseketens verbreken, waardoor stukken cellulose polymeer uit de structuur uitsteken en beschikbaar worden voor de cellulasen in de conventionele enzym mixen. Zo zullen ze bijdragen tot de afbraak en stimuleren ze tevens de activiteit van de klassieke enzymen. De succesvolle toepassing van LPMO’s als onderdeel van die enzymenmengsels vereist echter voldoende stabiliteit, om gedurende het verloop van de processen te overleven bij de vereiste temperaturen. Daarom omvatte het doel van dit onderzoek de verhoging van de thermische stabiliteit van een specifiek LPMO, namelijk Sterptomyces coelicolor  LPMO10C. Hiervoor werden de effecten van verschillende modificaties gemeten en geëvalueerd.

Het onderzoek

Als eerste stap werd een expressiesysteem op punt gesteld waarbij de correcte amino-terminus van het eiwit verzekerd werd en vervolgens geoptimaliseerd om voldoende eiwitopbrengst en zuiverheid te verkrijgen. Reeds verschillende onderzoeken hebben namelijk al aangetoond dat dit eiwit uiteinde cruciaal is voor de interacties die een koper-ion in het actieve centrum van het enzym fixeren. Dit koper ion is op zijn beurt onmisbaar voor de cellulose afbrekende activiteit. In dit onderzoek werd echter ook onderzocht wat het effect is van deze koper bindende amino-terminus op de stabiliteit. Uit deze testen bleek, dat bij het verbreken van de koper fixerende interacties van het amino-terminale einde van het enzym, dit niet alleen resulteert in het verlies aan activiteit, maar ook zal zorgen dat het enzym al denatureert bij een 7°C lagere temperatuur.

Vervolgens werden de meest flexibele regio’s van het enzym bepaald met röntgenstraal kristallografie en het algoritme van het B-fitter computerprogramma. Vertrekkende van de wetenschap dat de meest bewegende delen eerst zullen afbreken, werden deze meest flexibele regio’s aanzien als meest kwetsbare. Bijgevolg werden deze dan ook gebruikt als doelwit voor mutagenese, om ze door middel van extra interacties te fixeren op de meer rigide regio’s. Eén set gemuteerde enzymen betrof de introductie van zout bruggen, ionische netwerken en meer polaire residuen op het eiwitoppervlak, terwijl een andere set gebaseerd  was op de introductie van zwavelbruggen. Na productie werd de activiteit van deze mutanten getest door ze te incuberen met een cellulosesubstraat (cellulose behandeld met fosforzuur) en de afbraakproducten te scheiden, kwantificeren en analyseren (via High Performance Anion Exchange Chromatography met Pulsed Amperometric Detection). Verder werd via differential scanning fluorimetry (DSF) ook de smelttemperatuur gemeten. Hierbij fungeert de temperatuur waarbij het enzym maximaal ontvouwt, als maat voor de thermostabiliteit.

Resultaat

Bij het meten van de activiteit, bleek geen enkele van de extra ingevoerde interacties de oxidatieve splitsing van cellulose te verhinderen. Terwijl de activiteit op cellulose dus steeds gevrijwaard bleef, resulteerden niet alle mutaties in een verhoogde thermostabiliteit. In verschillende gevallen, was er geen noemenswaardig verschil te detecteren of resulteerde de extra interactie vermoedelijk in een verwrongen 3D-structuur van het enzym, waardoor de mutant reeds bij een lagere temperatuur denatureerde. Toch bleek de uitgewerkte methode succesvol, aangezien 4 mutanten na introductie van een extra zwavelbrug resulteerden in een verhoogde smelttemperatuur (2 keer +3 °C, 1 keer +5 °C en ook 1 keer +9° C). Hiervan werden vervolgens combinaties van de verschillende mutaties toegepast om te controleren op cumulatieve effecten naar stabiliteit toe. Terwijl sommige combinaties van mutaties elkaar teniet deden, bleek één combinatie te resulteren in een verbetering in stabiliteit die de exacte som was van de verbeteringen van de individuele mutaties.

De bijdrage aan een duurzamere toekomst

Uiteindelijk werd 1 finale combinatie van twee mutaties gemaakt, waarvan de temperatuur waarbij het denatureerde verhoogd werd van 51°C voor het wild type naar 63 °C voor de mutant. Hiermee is dit onderzoek voor zo ver we weten, het eerste onderzoek dat resulteert in een verhoogde thermostabiliteit van een LPMO. Hoewel verdere testen en optimalisatie vereist zijn voor de succesvolle implementatie van LPMO’s in de cellulose afbrekende enzymenmengsels, zou dit resultaat een belangrijke bijdrage kunnen leveren aan de ontwikkeling van een rendabel proces om van cellulose uit plantenafval een duurzame hernieuwbare grondstof te maken.
